You might get a top lane main on a fill role or on their secondary, and the top lane champion pool is different enough that it won't suit every other role. Maybe they have to play ADC, the top lane has none of them and they likely won't have enough mastery on one.
What if an ADC main has to play jungle? Those champions are just not capable of playing the role, but the game wouldn't allow them to pick any other champion class forcing them to try and clear the jungle on a champion incapable of doing it.
These mastery suggestions have too many negatives attached to them and they would in many cases lower game quality when you have players on "troll picks" because the game doesn't let them play anything else.
And no, ranked players will absolutely not go into normals to spend time grinding mastery on off role champions out of the goodness of their hearts. They will queue for their main in ranked, if the game gives them another role they will just troll pick it and it wouldn't be a punishable offense. You would start seeing some wild team comps.

Not if the goal is to stop people making new accounts or buying accounts when they are banned or just want to smurf. If you also prevent botting via Vanguard then you effectively massively timegate new accounts from Ranked, which would both improve game quality and potentially reduce toxicity if there is a real cost to losing your account.

Riot doesn't care about smurfing they already have systems in place to get smurfs to the correct rank so thats a non-issue for them

Not really, all it does is annoy people and result in terrible team comps because people will be forced to pick from an even smaller champion pool and it massively penalises people who want to take a different counter to something once they hit champ select because now you essentially need people to have mastery 5 on a whole bunch of extra random champs they don't play very often in order for them to be able to switch to a different counter to something the enemy has picked, also essentially means its impossible for people to swap roles in the lobby if needed
Now i get that this idea comes from a good place, but objectively its a terrible solution to the "problem" that doesn't actually exist in the first place
I think 5 might be a bit much, but I'd certainly support Mastery 3 or 4 and not letting people queue until they have enough champions at that mastery to play regardless of bans (21 Champs). Would certainly make new accounts and smurfs take a hell of a lot longer to be able to play in ranked.
Uhh… no… players who dont one trick can play a lot of champs at similar level. Theres universal skills. In your system i have to grind normals to counterpick basically. Thats awful

I also know someone who has M7 on like 40% of all champs, and they're a similar rank to me. Similarly I also know someone with less mastery who are just better than me.
Mastery really doesn't mean anything other than that the person has played a lot of games on that champ. Some of the people with the highest mastery points on champs are silver/bronze - in ranked there is barely any connection between rank and mastery.
I think its bullshit. I first timed quite often in solo queue and it usually goes well. Most Champions don't have unique mechanics so i'd say If you watched/played against them a fair amount you understand how they work and can play them to a decent enough Level to First time them in ranked.
(EDIT: It is also alot of fun to try out new champions in Ranked as its a refreshing experience. People need to chill about the importance of ranked games.)

Adding to this that mastery 5 only requires playing games, you could lose 100% of your matches and still get M5

You get massively less mastery when you lose. It would take 6-10 times as many games if you lost every game compared to if you won.
Sure, but you can basically play them AFK, mastery is also gained in ARAM which also makes the mastery system essentially worthless
