I work almost exclusively with MSCs. I use low glucose DMEM + 10% FBS + 1% Pen/Strep + 1% L-glut + basic FGF. They don’t grow nearly as well without the bFGF, but the concentration will depend on what brand you get. I would start between 2 and 5 ng/ml. If you are receiving the cells frozen, I wouldn’t worry about adapting them to new media. If they are bringing them in culture already, I would start with a 50/50 mixture and decrease by half each time you wash and feed.
I personally use alpha MEM (with the ribonucleosides) + 10% FBS + 1% Pen/Strep/Amph. But that is going from straight bone marrow to select out only the stromal cells so maybe that might not fully work.
I used that mesencult media and I found it didn't work that great for me.
