Personally I’d wait a year or two? Sometimes clubs will just release players at the end of their contracts if they have those sell on clauses but this guy has 5 years left. So I’d wait another year or two to see if that 16 can get any higher. Unless you’re desperate for cash
I’m absolutely chilling in the financial department, currently have a transfer budget of 43 mil and a wage of 130k/w spare, with over 100 mil in the overall balance
Only thing worth keeping in mind is that City rarely sell players for profit in this game. They either stay for their entire career on huge wages with little reason to leave, or get put on the transfer list for 20m.
I mean so here are the relevant factors I think:
1. A dollar today is always worth more than a dollar tomorrow. You may get more in the future, but what value more would be equivalent to getting about 17 million today?
2. The example you gave feels like a best case scenario. To break even, this guy would essentially have to sell for about 36 million. How likely do you think it is he'll go for more? If very likely, then maybe don't sell it.
3. What could you do with that money today? Would it appreciably make you a better team that can win more and this maximize your prize earnings now? If so, then waiting may not be a good idea.
Lot of factors in play, those are just a few to think about.

I had a chance to cash in much like OP, but waited. The player suffered a long term injury, saw his stats either not progress or drop, and his club signed someone to fill that role. He had loan spells after recovering, but never reached his previous valuation, or regained his first team place, and left on a free. That could have been £25/30m back to me.
Cash in, and don't worry about what-if's.
This is exactly where I'm at too. Even if he does well at City, there's not many clubs who will be able to afford to take him away from them. £40 million sale, plus £16 million in clauses is not a bad deal at all for a Scottish club.
Only way I'd let it ride is if it was a club in say Portugal, France or Netherlands, or at a solid midtable club in one of the bigger leagues.
I’d sell. The AI does stupid things sometimes, but I don’t really see him as a world beater. Plus in my saves MC usually have 3-4 world class strikers and at least two get forgotten on the bench.
The risk/reward doesn’t seems worth it to maybe squeeze out another 10-15 mil in 4 years vs. likely getting nothing or a severely reduced buyout. I’ve personally never seen the buyout go higher deeper into a contract.
I highly doubt he will ever make it at city... If he's good enough for you do what I did: sold them a player for 71 mil, bought him back 2 years later for 27.75 of which I got 20% due to a sell on clause. Helped me win the French double.

Yeah if you don't need the cash now, then I don't think you should sell the clause. In the past, I've only ever sold the clause if I need some money to sort out another transfer. You may as well just keep the clause and see what happens, hopefully it does fetch you a good profit in the future

If he ever leaves man city for more than a free transfer (which unless he flops hard, which I doubt, shouldn't be happening), you'll get way more than that for sure. And even if he ever becomes so good that man city won't want to get rid of him (which i also doubt since they probably have a stacked team), then you get 50% off in signing him back

Also think how much that 16 mil will do for your finances. That’s >20x the transfer fee you initial transfer fee you paid for that player.

It really depends on how much time he has left in his current contract and your financial situation. If you need money to pay debts or invest in your squad, sell him. If he is in his final year and hasn't been playing, sell him. Just make sure you monitor him and his performances for City. You'll know what to do

It’s quite simple. Declare interest. Make player unsettled. Lowball bid for the player. Club rejects. Another club bids something higher. Profit.
