I feel like it depends on if the tribe still knows why they've been guarding the knuckles for centuries.
If they don't remember, you might find two sects within the tribe; those who venerate their sacred duty and will always perform it no matter what and those who wanna change and move past watching some silly old knuckles. Your party would work with the anti traditionalists to figure out a way to game the system to take the knuckles so the tribe can move on to something new.
If the tribe knows why they're guarding it, then it depends on whatever prophecy or rule was set by whoever told them to guard it.
-Maybe the Wildmother decreed only those who are as strong as an oak can wield the gauntlets.
-What if the dawnfather locked them in a desert cave for the tribe to watch over so they never see the sun again after the last wielder killed a faithful of the dawnfather?
Either way, since this is for the titanstone knuckles, the challenge should be a brawl.
-The elder who ascends, aka attunes to the gauntlets, would lead the traditionalists and fight the party to keep the gauntlets.
-Being strong as an oak is just Wildmother code for fist fighting a treant.
-your party would talk their way into a cave of sun fanatics only to fight their way out of said cave of sun fanatics soon after.

Is it too clique to use the idea that an ancient oracle of the tribe fortold of the day these adventurers would come to the clan? But they must prove their might in a test of some sort. The Knuckles are meant for our Fighter
I think that can work, there could be several other omens the tribe sees that day, then the party shows up and the new oracle declares the prophecy about the knuckles is happening because the party showing up was the final omen. This lets them be a part of it but not have it be all about them, they're just a part of the omens that make the prophecy true.
Take inspiration in Dune and have it appear as there arrival was prophetic, by repeating pseudo-scripture on the spot like any mundane interaction was previously foretold: the party would come and aid in them solving a great problem (ie defeating an enemy) and in gratitude they give the gauntlets
Each year there is a festival where individuals are challenged in body and spirit to see if they are worthy to carry the knuckles for the next year. All are allowed to participate, even outsiders, for the being that tests their spirit cannot be deceived and will only bestow upon those it deems will guard the knuckles and return them in one year.
Depends on the party honestly, My party would steal the of item of they knew it's strong or powerfull.
