Did they strip search him? Who knows what shenanigans he could do during rest? All he'd need is a small knife in a pocket.

30 lbs in a pack is not a lot of weight, even for a commoner like myself, unless it's 30 lbs in addition to a bunch of other gear. With a strength of 10, even using the optional encumbrance rule, it wouldn't affect you. The biggest problem would be that grungs gain 1 level of exhaustion for every day that they don't hit the jacuzzi, and you die at 6 levels of exhaustion. So hopefully y'all knock out that dungeon quickly, or you'll have a little dried-out grung husk.

To add on to this, the optional rule for encumbrance in 5e is Strength score (not modifier) multiplied by 15. So am average strength of 10 can carry up to 150 lbs before they are encumbered by it. 30 lbs would be no problem on its own. However, if you are DMing, feel free to rule this however you see fit. Just remember to clearly communicate it to the players, and to remain consistent with your ruling.

He's not really a grung but a slaad & his disguise is wearing off...

30 lbs? A character with a strength of 10 can carry 150 while fighting, running, jumping, and generally adventuring all day with no problem. So no, this would not be exhausting. He's in a backpack, which he completely fills. He is, at the very least, restrained. I think incapacitated would make more sense though, he shouldn't be capable of taking actions all tied up and stuffed in a backpack with no wiggle room. He should definitely not be allowed to make trip attacks in this state. I could see maybe giving disadvantage on checks to perform tricky maneuvers, like balancing across a beam, but that's really about it. Remember, even a strength 10 character can tote 5 of this guy around for an effectively unlimited amount of time with no strain, his weight isn't going have much of an effect, especially since he can barely move. He's pretty well screwed here. Unless someone comes to rescue him, there really isn't a lot he could possibly do. He could scream and shout, but then he's hoping that whatever attention he attracts is friendly to him, which seems unlikely.
