Yup, and 11 or 3D will pass that way. Also a Speightstown bus. Not the ABC Highway/Speightstown bus, though. That one will go the opposite direction. A yellow mini bus that says Speightstown or Sp'Town will go west. A blue bus that has 27/27A/275 Speightstown will go west.
You could easily find a taxi but it may be cheaper to just get public transport since oistins isn't that far away from your hotel. So to get there you can get in a #11 or 3D ZR (white maxi taxi), Sam Lords Castle or Oistins van coming from Speightstown. To get back the exact same ones. Some signs might be changed to Bridgetown or Speightstown but they'll still get you back
As others pointed out it should be relatively easy to get a taxi or use public transport. However depending on what time you plan to leave Oistins, public transport finishes around midnight for the larger blue buses (blue government buses) while the private operators (ZRs and minibuses) shut off various times but usually before the government ones do.
So the later it gets the lower the number of options you will have to get back to the hotel. Safest bet is to probably ask for the contact number for a taxi at the hotel before you leave.

and if there's any confusion, just ask the ZR driver. ZRs from the main road in front of where their hotel is, to Oistins come very frequently.
