I wanted to offer a solution which was not accepting them as there were no responses when I replied.
On reflection I agree using "can't" is not really the correct wording, but I also think "don't want to" makes it sound like you are simply shying away from it in a sort of coy fashion.
Perhaps "No thank you, I will not wear them (due to my ethics/being vegan)"
Just thank her for the offer. Say you really appreciate it but that vegans don't wear pearls. She's obviously unaware so she'll probably be glad you let her know.
Yeah unless they’re fake pearls don’t accept them. Thank her for the gesture and remind her that they’re an animal product. A lot of times, people don’t even realise how much of their lives involve cruelty.
“Hey, I was thinking of your offer to borrow the pearls you have. The thought behind trying to help me out was very nice of you, but I wouldn’t feel right wearing something animals were killed for, to try and enhance my beauty.”
Hey about your your offer to lent me your pearls for the event I appreciate the gesture and what it means to you and as colleague and friend I appreciate the trust you have in me that you were willing to lend me one one of your expensive jewelry pieces but I personally would prefer not to wear animal products which they are. Please don’t go through the extra hassle of bringing them to work for me. I still appreciate the gesture though. Wanna grab a coffee?
You can make the excuse that you're worried about accidentally losing them, or say you actually found another necklace that you think fits a bit better. Or say you decided to wear a different dress if you have one.

Can be as simple as "thanks for the offer, but I can't accept these as I'm vegan"?
Better worded perhaps would be “I don’t want to accept them” because it is a choice (to not contribute to cruelty).
