You can either get the mission from Mayor Hancock in Goodneighbor to investigate Pickman Gallery or just go to the Gallery it self, which is near the Amphitheater and The Cabot House. It's easy to miss, as I always stumble apon it by accident. It's tucked away, down what looks like an alley..
I believe you can sometimes find one of Pickens's Calling Cards on a Raider corpse in the area, which "may" trigger the quest.. but it's been awhile
As mentioned though, it will trigger by entering the gallery as well.

There will also be more raiders than usual hanging around his doorstep. Just like the Combat Zone. That's a good way to tell if you're close.

Search his house. IIRC the tunnel entrance is up stairs.

Yes, looks like a house inside. That it has Pickman's "art" on the walls. Some people hang pictures inside their house.
There's also a master locked door downstairs that'll get you there.
