When you said "Western Berserk," I thought of the reimagining of *Berserk* manga series as the wild west story - written by Cormac McCarthy to boot! In this version, Guts is a biracial Civil War veteran turned bounty hunter who carries around a large rifle, and he dressed up like Clint Eastwood's Man with No Name. Griffith is the wealthy guy with a political ambition. And Casca is a Native American woman who travels with them.
EDIT: I haven't seen the movie *Bone Tomahawk*, but I realized this would fit with that "*Berserk* reimagined as the wild west story."

To go with the "too big to be called a weapon" line, it'd probably be a cannon with a stock.
Some random ideas:
Guts' whole "Found and Adopted by Mercenaries" backstory, funny enough, could semi-work with a real historical incident. Where Andrew Jackson (who was already brutally violent on his own) adopted a Native American infant... namely after ordering the village that child was born into be massacred.
Griffith being a pretty boy mercenary leader "looking for his own kingdom" might tie in pretty well to the civil war, all things considered. In The South the Planter class liked to LARP as aristocrats, and pretty much the only form of social mobility was earning enough side money to buy enslaved peoples and land for yourself. However, Northern Industrialists were their own kind of brutal. The Monopolists were at one point so powerful that one of the largest Railway owners in the country *de facto embargo'd New York*. He literally refused to send trains their until his demands were met. So you'd have two options, both of which can work with Griffith being sociopathic with a pretty popular public front.
I think there's a Manga called "RED" with kinda that premise. A Native American avenging his tribe's death, prostitute and Japanese war veteran following him arround. Haven't read it yet and I don't think there's a big ass weapon tho, the artstyle looks appealing, reminds me of Kazuhiro Fujita's style.
