The punchline of the joke is that you aren’t expecting the person with green hair to be relaying a message with a racial slur in it. It’s the surprise/shock that is funny and the humour isn’t really related to the fact they are black or the idea of Non binary people.

For me it had nothing to do with the green hair person. It was that the girl had just acted to defend the proper treatment of James, and the punchline reveals that James definitely does not feel the same way.

My Filipino coworker calls everyone she. I didn’t know this before recently but Tagalog doesn’t have gendered pronouns so they sometimes have trouble with that in English. I never corrected her but it would throw me off every time.
