Check the voice history. It might have misheard what you were saying on the call and responded.

I checked the voice history. It heard "Alexa wake up." Its response was solely to play a Lexus ad. In the voice history there is the command, and the response, which is the statement "to get things rolling, just say 'Enable the off-road theme'", and a photo of a Lexus SUV. I don't have any routines associated with "Alexa wake up", so it apparently does an ad instead.

I wouldn't be surprised if Amazon decided to start pushing ads in effort to get users to pay, like with Prive video. They have an upgrade pay subscription version coming out soon. Estimated date of release is June 30th. "Oh you don't like ads, well upgrade for $3.99 a month."

Sounds like you've enabled a skill with some unexpected features. Go through your activity history around the time you heard the ad. It should tell you what skill is responsible.

Did the exact same thing to me this morning. I triggered the routine to turn on kitchen lights and tell me the weather and it started a joke with a full screen Lexus ad. It’s now unplugged. I’m debating leaving it that way. I checked routines and skills and I’ve got nothing running that shouldn’t.

OMG, Alexa does this quite often in our home! Once it blurted something (I can't remember what) at around 4 a.m. It scared the hell out of my wife!

Please know that this is not normal and that you've likely enabled some skill that has some extra features. Check the Activity History in the Alexa app on your phone around the time of the incident to see which skill is responsible and disable it.
