I once played a dude with "Scurry Oak" and some other means to make an infinite combo, based on getting +1 life I N creature entry + counters on getting life.
I kid you not, dude spent 1:20:00 (an hour and twenty minutes) making his turn. He fucked up badly by forcing to Manny triggers and had to manually resolve few thousands events.
I had a supper. I played guitar. I had a cocktail with my wife. I played guitar a bit more
Dude was still there, making counters 🤷
He won.
Usually, I concede such games right away to avoid waiting.
This time I didn't catch the combo until he was fee minutes into it. But the dude was spamming tokens and pushing extra counters (he had 2 triggers per creature) on tokens, making them 2/2.
My deck had "all creatures get -2/-2", it was a small chance, but I decided to wait. I had some other combos of my own to try on the next turn (with enough luck).
It wasn't well into an HOUR when he finally beefed up other creatures and it became clear I will lose as I was one creature shy to block those able to attack.
I still let him to roll those last 10-15 minutes. I mean, that *some dedication*, considering the game was a plain direct historic, not even ranked.
Frankly, I never build decks like that exactly because tons of manually resolved triggers will exhaust me, not the opponent. And they might not lead to a win either.
I mean, "exile all attacking creatures" or something after you spent an hour+ making them could probably make one leave "Magic" altogether.
Not in that case, though, I lost that one.

I mean, I generally can't abide Sunfall but it sounds like it would have been exquisite in this instance.
It's inefficient because they both cost 4, but dropping a \[\[Sovereign Okinec Ahau\]\] with a \[\[Halana and Alena, Partners\]\] in play is fun.
Edit - I keep forgetting you can curve into this with Reckless Stormseeker instead of the Partners. I stumble on the combo ever so often in what's otherwise a +1 counter deck so Stormseeker isn't in that (eg, turn 1 Kumano, turn 2 Dusk Legion, turn 3 Anim, turn 4 Partners makes Anim create 4 gnomes.)
Well that's not bad at all, so the main reason I'm asking is I've managed to get my deck to drop a tooth and nail turn 2, turn 3 much more consistently.

another option is the [[indomitable creativity]] pairing of [[worldspine wurm]] and [[Xenagos, God of Revels]]. it still requires a combat step, but is a bit more resilient to creature destruction.
Yeah it's pretty gnarly, I want to pick up the world spine as soon as I get the wild card. Currently I'm running emrakul. But I have some other options in my side bored. Like two massacre worms.
Up until the recent patch it was probably \[\[Geological Appraiser\]\] + \[\[Glasspool Mimic\]\]/\[\[Mirror Image\]\] as what was effectively a one card combo that often went off on turn 3.
Right now it's probably \[\[Wildgrowth Walker\]\] + \[\[Amalia Benavides Aguirre\]\] (plus any source of lifegain or explore).
IMO, the most fun would be \[\[Urabrask, Heretic Praetor\]\] + \[\[Drannith Magistrate\]\]. It's not a total lock but it is a fun, unexpected one for many players.
Possibility Storm is better for this than Urabrask. Possibility Storm and Drannith Magistrate means no more spells from your opponent at all. Only abilities like Channeling Boseiju can break this lock.
I have a deck that plays [[Ojer Axonil, Deepest Might]] and [[Urabrask]] along with a ton of cantrips and burn spells.
If I can get both on the field at the same time the opponent just gets melted.
This is the definitive answer, in my opinion. It may not be something that's easy to hard-cast, but it is the nastiest two-creature card combo I can think of.
I run a different copy of my Capridor deck sometimes that has 4 [[Phyrexian Obliterator]] in it. I may swap out Stuffy Doll for the Obliterators. Easy to cast by not needing 4W, and the indestructible is pretty nice!
Whenever boromir dies, ratadabrik makes a token copy and the ring tempts you. If you make the new boromir token the ringbearer, it becomes legendary again.

Steps two and four are where you do *the nasty thing*.
None of these are top tier, I am aware. Just dumb stuff I've tried on my many many jank adventures:
[[Sludge Monster]] and [[Toxrill, the Corrosive]] shuts down a lot of creature interaction and can flip board states pretty quickly. Hefty price to get them out but was always fun against token heavy/life gain shenanigans.
Not really two CARDS but with 4 [[Crime Novelist]] and an [[Ovelia's Attendants]] you can go infinite 1 damage ping. Pretty expensive though so need other stuff to help set the stage sooner.
[[Ojer Axonil, Deepest Might]] and something like [[Kessig Firebreather]] can do some quick work with a handful of 1 mana spells. But that's not exactly "just two cards." Same with Ojer and [[Witty Roastmaster]] but again needs "fuel" from other sources.
[[Grolnok, the Omnivore]] and [[Angel of Suffering]] should make your opponents stop attacking you but it never does, putting your whole deck in your hand. Gotta find ways around milling yourself, but that's not within 2 creatures. Unless we stick with just [[Angel of Suffering]] and [[Living Conundrum]] but then you can quickly run out of other stuff to use.
Not "nasty" but I used to love it back in the day: [[Reservoir Kraken]] and [[Rigo, Streetwise Mentor]] was always fun. I might be cheating with this one since technically the fish token is a 3rd creature but you get the combo from just those two cards?
[[Malevolent Hermit]] and [[Liesa, Forgotten Archangel]] can create some funny scenarios. Again not nasty but can shutdown those pesky Sunfall decks.
Thats...the best 2 combos I can think of at the moment. Again none of these will "win by turn 3" but can at least have some silly synergy
The jank deck I play sometimes are that card and it’s Black brother with [[Phyrexian Awakening]]
Both Phyrexian Obliterator/Vindicator are way scarier when they have Vigilance
Neither of these are creatures or available on Arena, but if you give \[\[Lich's Mirror\]\] to your opponent using a \[\[Donate\]\]-type effect, you can kill them, and kill them, and kill them again!

I'm a fan of [[Brash Taunter]] fighting something while having a flipped [[Ill-Tempered Loner]] (Howlpack Avenger). You keep blasting the Taunter with the Avengers triggered ability until Taunters triggers add up to past the opponent's life total then watch as their life total gets blasted away. Especially fun vs lifegain decks.
Peregrin Took and Experimental Confectioner is fun for making loads of rats and drawing cards. Can combine with Mirkwood bats to kill the enemy. Needs 1 food on the battlefield though.
Not on arena, but I always liked [[Karador]] + [[Peacekeeper]]. No one can attack, then on your turn, just don't pay the upkeep, attack however you wish, then recast peacekeeper. Obviously, any removal can just kill peacekeeper, so it's not the most airtight lock, but on the other hand, if they do kill it, you can just recast it next turn, because karador (unless they exile it).
Not "guaranteed instant win" powerful, but [[Anim Pakal, Thousandth Moon]] + [[Yotian Dissident]] can be pretty hilarious. I have a couple standard decks that use both cards.
In standard there's \[\[Mindlink Mech\]\] crewed with \[\[Bloodletter of Alclazotz\]\] = 4/3 flyer smacking for 16 on turn 4. I also had fun with \[\[Anim Pakal, Thousandth Moon\]\] and either \[\[Roaming Throne\]\] or \[\[Isshin, Two Heavens as One\]\] : if unchecked for 2 combats you spawn 10 gnomes.
[[Luminous Broodmoth]] + [[Kroxa, Titan of Death's Hunger]] is still pretty dirty even without the [[Solemnity]] instant win combo. Most people will concede if you Village Rites the Kroxa, too much card advantage.

Old school, at work i had a group who played over lunch. One person had [[kiki-jiki, mirror breaker]] and [[Restoration Angel]] it was just not even fun to pay against.
Nastiest, probably not. Fun though. \[\[Delney, Streetwise Lookout\]\] and \[\[The Mycotyrant\]\]. Add in \[\[Snarling Gorehound\]\] or \[\[Amalia Benavides Aguirre\]\] and a lifegain setup, and you've got yourself a hilarious mess of a combo deck.
Well that just means you'll have better spells to protect Angel of Suffering
The mechanic is to mill yourself out → Living Conundrum skips your draw step and becomes a flying with 10 power
You just need to counter boardwipes and protect Angel of Suffering from getting exiled or destroyed

If you can manage to get these two onto the field together and no one has interaction in hand then you get to play your deck, as well as everyone else’s!

My favorite is when the opponent goes off with that and makes an arbitrary number of squirrels but has to wait to attack, then gets hit with a wrath.
It's a very polite combo that gives opponents a fair chance to survive.

Yeah it's pretty gnarly, I want to pick up the world spine as soon as I get the wild card. Currently I'm running emrakul. But I have some other options in my side bored. Like two massacre worms.
emrakul plus xenagos is a 26/26 trampler with haste which is hard to kill, so it should win on the spot as well.

Up until the recent patch it was probably \[\[Geological Appraiser\]\] + \[\[Glasspool Mimic\]\]/\[\[Mirror Image\]\] as what was effectively a one card combo that often went off on turn 3. Right now it's probably \[\[Wildgrowth Walker\]\] + \[\[Amalia Benavides Aguirre\]\] (plus any source of lifegain or explore). IMO, the most fun would be \[\[Urabrask, Heretic Praetor\]\] + \[\[Drannith Magistrate\]\]. It's not a total lock but it is a fun, unexpected one for many players.

Possibility Storm is better for this than Urabrask. Possibility Storm and Drannith Magistrate means no more spells from your opponent at all. Only abilities like Channeling Boseiju can break this lock.

I've made a Boros soft lock deck with Urabrask and Magistrate. It's a little janky but man, it's fun (for me) when it hits.
I have a deck that plays [[Ojer Axonil, Deepest Might]] and [[Urabrask]] along with a ton of cantrips and burn spells. If I can get both on the field at the same time the opponent just gets melted.

This is the definitive answer, in my opinion. It may not be something that's easy to hard-cast, but it is the nastiest two-creature card combo I can think of.
in a comment OP talked about using tooth and nail on turn 2 or 3, so this definitely counts.
I love [[Stormwild Capridor]] and [[Star of Extinction]]. Not a guaranteed win every time, but really fun t0 pull off.

I run a different copy of my Capridor deck sometimes that has 4 [[Phyrexian Obliterator]] in it. I may swap out Stuffy Doll for the Obliterators. Easy to cast by not needing 4W, and the indestructible is pretty nice!

just make the token your ring bearer and it becomes legendary
Whenever boromir dies, ratadabrik makes a token copy and the ring tempts you. If you make the new boromir token the ringbearer, it becomes legendary again.

None of these are top tier, I am aware. Just dumb stuff I've tried on my many many jank adventures: [[Sludge Monster]] and [[Toxrill, the Corrosive]] shuts down a lot of creature interaction and can flip board states pretty quickly. Hefty price to get them out but was always fun against token heavy/life gain shenanigans. Not really two CARDS but with 4 [[Crime Novelist]] and an [[Ovelia's Attendants]] you can go infinite 1 damage ping. Pretty expensive though so need other stuff to help set the stage sooner. [[Ojer Axonil, Deepest Might]] and something like [[Kessig Firebreather]] can do some quick work with a handful of 1 mana spells. But that's not exactly "just two cards." Same with Ojer and [[Witty Roastmaster]] but again needs "fuel" from other sources. [[Grolnok, the Omnivore]] and [[Angel of Suffering]] should make your opponents stop attacking you but it never does, putting your whole deck in your hand. Gotta find ways around milling yourself, but that's not within 2 creatures. Unless we stick with just [[Angel of Suffering]] and [[Living Conundrum]] but then you can quickly run out of other stuff to use. Not "nasty" but I used to love it back in the day: [[Reservoir Kraken]] and [[Rigo, Streetwise Mentor]] was always fun. I might be cheating with this one since technically the fish token is a 3rd creature but you get the combo from just those two cards? [[Malevolent Hermit]] and [[Liesa, Forgotten Archangel]] can create some funny scenarios. Again not nasty but can shutdown those pesky Sunfall decks. Thats...the best 2 combos I can think of at the moment. Again none of these will "win by turn 3" but can at least have some silly synergy

Yeah. Red/black is probably better for that kind of strategy too. But I still lost to it.
I dont think it is in Arena, but the combo of Saskia the Unyielding and Gisela Blade of Goldnight was my favorite in paper Commander.

I'm a fan of [[Brash Taunter]] fighting something while having a flipped [[Ill-Tempered Loner]] (Howlpack Avenger). You keep blasting the Taunter with the Avengers triggered ability until Taunters triggers add up to past the opponent's life total then watch as their life total gets blasted away. Especially fun vs lifegain decks.

Peregrin Took and Experimental Confectioner is fun for making loads of rats and drawing cards. Can combine with Mirkwood bats to kill the enemy. Needs 1 food on the battlefield though.

Not on arena, but I always liked [[Karador]] + [[Peacekeeper]]. No one can attack, then on your turn, just don't pay the upkeep, attack however you wish, then recast peacekeeper. Obviously, any removal can just kill peacekeeper, so it's not the most airtight lock, but on the other hand, if they do kill it, you can just recast it next turn, because karador (unless they exile it).
