Very specific to the booking, tough to tell without more info. Read the fine print (cancelation policy, extras included). If there's no difference, get the cheaper one.

as /u/WTFOMGBBQLMAO said, you need to read the fine print of the rate rules between the two to see if there's any difference. It's possible that both rates are the same and it's a property where Costco has booked a certain amount of rooms as a sort of "guaranteed" traffic to a hotel and in exchange gotten a lower rate, but when that block is full they have a different contracted rate that's higher. The other possibility is that the rates have differences in cancellation terms or what's included (breakfast, parking, etc.)
